Microbiological Assay of Vitamins by Turbidometric Method
1. Aim
To interpret a vitamin bioassay standard curve and estimate a sample level from a valid turbidimetric record. The vitamin and measured concentration must be entered in the student record.
2. Requirements
Instructor-supplied standard and sample absorbance records, vitamin reference-standard certificate, blank and growth-control data, validated method, spectrophotometer record, USP <171> if vitamin B12 is assigned. Vitamin identity, organism, wavelength, medium, preparation, incubation, curve range, and acceptance rules are to be recorded from the assigned current monograph or approved college method in the student record.
3. Theory
A microorganism that requires a vitamin can show increasing growth over a useful standard range when that vitamin is supplied. Turbidity measured by an optical instrument acts as a growth signal. The blank, growth controls, and standard curve are needed to separate vitamin-related response from background. The unknown is interpolated within the validated range, then corrected for documented dilution. This is not a general formula for all vitamins; the selected vitamin monograph and local validated method control the assay.
4. Procedure
- Identify the assigned vitamin and validated method, then check the blank and growth controls.
- Record the standard concentrations and corresponding turbidity readings from the supplied dataset.
- Plot the approved response model and examine whether the standards meet its validity criteria.
- Read the unknown only within the accepted standard range and account for documented dilution.
- Report the result with units and an assay-validity statement; unresolved calibration details must be entered in the student record.

Assumed values for practice only: an assumed calibration table maps an assumed absorbance of 0.40 to an assumed vitamin level of 2 arbitrary units/mL. This illustrates interpolation only. The actual vitamin, valid curve, dilution factor, and concentration must be entered in the student record. Arbitrary units are not a release specification.
6. Output or final result
Report a vitamin level only from a valid assay curve and documented sample dilution. Record the supervised outcome here.
Teaching-lab safety: Use supplied readings or supervised approved organisms and reagents. No vitamin concentration or culture condition here is a preparation instruction.
This page is a supervised-activity guide; it does not report a completed case.
