Preparation of Syrup of Ferrous Phosphate IP
1. Aim
To study and, where an authenticated formula and trained instructor are available, prepare a teaching-lab sample of Syrup of Ferrous Phosphate IP. Record the calculation, controlled mixing stages, final volume, and appearance. The applicable IP monograph and local authorization must be specified in your college lab manual and instructor's method sheet. [1, 2]
2. Requirements
The East Point College of Pharmacy teaching manual names iron turnings, phosphoric acid, calcium carbonate, potassium bicarbonate, sodium phosphate, sucrose, cochineal, orange-flavored water, and purified water. The authenticated formula may differ, so the exact ingredients, grades, acid strength, quantities, final volume, and required tests are to be recorded from the assigned current monograph or approved college method in the student record. Apparatus includes a balance, measuring devices, suitable reaction vessel, water bath, stirring rod, filter, calibrated final container, label, and batch record. An instructor must specify a suitable ventilated work area, eye and face protection, acid-resistant gloves, laboratory coat, and spill response. [2, 3, 4]
3. Theory
This syrup is assembled from an iron-containing acid solution, other mineral ingredients, and a sweetened vehicle. The college manual describes an iron and phosphoric-acid stage, a separate carbonate and phosphate stage, and a syrup stage before combining the liquids. Contact between iron and acid can release flammable hydrogen. Carbonates reacting with acid can cause effervescence. Phosphoric acid can injure skin and eyes. Therefore the reaction must remain open to safe ventilation, away from flames and ignition sources, under a trained instructor's written procedure. [2, 4]
The manual's formula table and stepwise interim volumes are not fully consistent, and the original IP monograph was not directly available. Treat the quantities, order of addition, heating conditions, filtration, and standing period in the student record. A clear or colored syrup does not establish iron content, stability, or compendial quality. This is not a patient-use preparation. [2, 3]
4. Procedure
- Ask the instructor to authenticate the IP formula, edition, target volume, complete sequence, acid concentration, and local chemical-risk controls. These must be specified in your college lab manual and instructor's method sheet. If they cannot be established, perform only the calculation and flowchart review, with no wet-lab reaction. [2, 3, 4]
- In the approved area, inspect ingredient labels, protective equipment, ventilation, balance, glassware, and waste containers. Keep flames and other ignition sources out of the work area. Record the safety check here. [3, 4]
- Calculate each ingredient amount from the authenticated formula and assigned final volume. Have the calculation and independent check recorded before dispensing any acid or iron. [2, 3]
- Under direct supervision, prepare the iron and phosphoric-acid portion exactly as the verified local method states. Control addition and temperature, keep the vessel unsealed, and avoid exposing personnel to splashes or evolved gas. The addition order, water amount, and heating conditions must be specified in your college lab manual and instructor's method sheet. [2, 4]
- In a separate vessel, prepare the specified carbonate and phosphate portion according to the verified method. Allow room for effervescence. The quantities and order must be specified in your college lab manual and instructor's method sheet. [2]
- Prepare the sucrose vehicle and any specified color and flavor components by the approved method. Allow hot liquids to cool before final-volume adjustment. The heating, filtration, and cooling instructions must be specified in your college lab manual and instructor's method sheet. [2]
- Combine the prepared portions in the authenticated sequence, with stirring and appropriate splash control. Clarify or filter only as the written method specifies. Do not seal a vessel while gas evolution is possible. [2, 4]
- Transfer to the specified calibrated container and add purified water to the verified final volume. Record appearance, sediment, final volume, any waiting period, and any further filtration here. [2, 3]
- Label as a supervised teaching sample, not for patient use. Record batch details and follow the institution's acid, iron, and contaminated-material disposal procedure. Storage and disposal details must be specified in your college lab manual and instructor's method sheet. [3]
5. Calculation, graph, or images as appropriate
| Item | Authenticated master formula | Scaled quantity | Actual record |
|---|---|---|---|
| Iron and phosphoric acid | Use authenticated formula | Record here | Record here |
| Carbonate and phosphate ingredients | Use authenticated formula | Record here | Record here |
| Sucrose, color, and flavor components | Use authenticated formula | Record here | Record here |
| Purified water to final volume | Use authenticated formula | Record here | Record here |
| Appearance, sediment, and final volume | Criteria record from the assigned current monograph or approved college method | Not applicable | Record here |
Worked scaling method: If the authenticated master formula specifies m grams of an ingredient for V mL of finished syrup, the amount for an assigned v mL batch is m × v/V grams. For a batch one-fifth of the authenticated master volume, the ingredient amount is m/5. Water remains sufficient to the final volume, not a separately added full volume. The actual m, V, v, and each numerical result must be recorded here. This arithmetic does not resolve the college manual's inconsistent interim volumes. [2]
6. Result
Record whether a wet-lab batch was authorized, the authenticated formula, calculated quantities, final volume, appearance, and deviations here. If no authenticated method was available, report that only a paper exercise was completed. Do not claim IP compliance or patient suitability from appearance alone. [2, 3]
