Preparation of Syrup of Ephedrine Hydrochloride NF

1. Aim

To document the calculation and supervised teaching-lab preparation of Syrup of Ephedrine Hydrochloride NF from a verified source formula, then assess the sample's appearance and final volume. The applicable NF edition, official formula, and local authorization must be specified in your college lab manual and instructor's method sheet before the active ingredient is handled. [1, 2]

2. Requirements

Ephedrine hydrochloride, a verified simple-syrup vehicle, and any other ingredients required by the approved NF formula; a calibrated balance, weighing paper or vessel, beaker, stirring rod, suitable graduated device, clean final container, label, and record sheet. The official NF monograph, ingredient grades, exact ingredient amounts, target final volume, bottle type, and any required filter must be specified in your college lab manual and instructor's method sheet. Wear a laboratory coat, eye protection, and gloves; follow the institution's controls for access, inventory, spills, and disposal. [2, 3, 4]

3. Theory

A medicated syrup contains an active ingredient dissolved or dispersed in a sweet syrup vehicle. The East Point College of Pharmacy manual lists ephedrine hydrochloride and syrup as the components of this named exercise. It reports a teaching formula, but the historical NF monograph and its edition were not available for direct comparison. Record the historical NF edition, strength, and any additional requirements from an authenticated monograph. This classroom formula does not establish current compendial suitability. Do not infer a clinical dose from a classroom formula. [2, 3]

The active ingredient is measured by mass while the finished preparation is brought to the specified final volume. Mixing a measured powder with a full target volume of syrup would overshoot that volume. A clear sample is only an appearance observation; it does not prove potency, uniformity, stability, or fitness for dispensing. [2, 3]

4. Procedure

  1. Have the instructor or responsible pharmacist supply the authenticated NF formula, edition, assigned final volume, and local authorization. Record each authenticated detail on the batch sheet before proceeding. If these are absent, complete only the paper calculation and diagram review; do not handle ephedrine hydrochloride. [2, 3]
  2. Review the ingredient identity, safety information, local access and inventory record, and the written procedure. Prepare a clean work area and wear the required protective equipment. Record the local control check here. [3, 4]
  3. Calculate the required ingredient mass from the verified strength and target final volume. Record the formula units and have the calculation checked before weighing. The values must be specified in your college lab manual and instructor's method sheet. [2, 3]
  4. Under supervision, weigh the approved amount of ephedrine hydrochloride with a calibrated balance and record the actual mass. Keep the ingredient in the controlled work area. [3, 4]
  5. Place a portion of the verified syrup vehicle in a clean beaker, add the weighed ingredient, and stir until the powder is fully incorporated according to the approved method. The starting syrup portion and mixing conditions must be specified in your college lab manual and instructor's method sheet. [2]
  6. Transfer the mixture to the specified calibrated container. Rinse the beaker with small portions of syrup if the approved method calls for it, and add syrup to the target final volume. Do not add the entire target volume before the active ingredient. [2]
  7. Inspect the sample for visible particles and uniform appearance. Record actual final volume and any loss or deviation. Filtration, if required by the authenticated formula, must be specified in your college lab manual and instructor's method sheet. [2, 3]
  8. Label the container as a supervised teaching sample, not for patient use, with ingredients, strength only after verification, batch record, and date. Follow the institution's storage, inventory reconciliation, and disposal procedure; these local details must be specified in your college lab manual and instructor's method sheet. [3, 4]

5. Calculation, graph, or images as appropriate

Formula or observation item Source or planned value Actual record
Authenticated NF edition and formula Use authenticated formula Record here
Assigned final volume and unit Use authenticated formula Record here
Ephedrine hydrochloride strength and unit Use authenticated formula Record here
Calculated and actual ingredient mass Use authenticated formula Record here
Syrup vehicle added to final volume Use authenticated formula Record here
Final appearance and container Use authenticated formula Record here

Worked calculation by symbols: If an authenticated formula specifies C grams of ephedrine hydrochloride per V mL of finished syrup, and the assigned batch volume is v mL, required ingredient mass = (C ÷ V) × v grams. For example, a batch one-tenth of the authenticated formula volume requires one-tenth of each scalable ingredient quantity. The source formula's C, V, assigned v, and numeric result must be recorded here. Check whether any ingredient must be added by mass rather than scaled by volume before using the result. [2, 3]

6. Result

Record the verified formula, calculated and weighed ingredient amount, final volume, appearance, and inventory reconciliation here. Record any discrepancy and do not represent the sample as NF-compliant or suitable for dispensing without confirmation of the actual monograph and required quality checks. [2, 3]
