Sterilisation of Glassware and Preparation of Media

1. Aim

To document the preparation and verified sterilisation of approved teaching-lab glassware and medium. Follow the college lab manual and instructor's method sheet for the medium formula, validated load cycle, and biological indicators.

2. Requirements

Instructor-approved glassware and commercial or specified medium, water, balance, pH meter if prescribed, labels, heat-safe containers, sterilizer, load records, protective equipment, and waste route. Follow the college lab manual and instructor's method sheet for ingredients, pH, validated cycle, fill volume, and quality controls.

3. Theory

Media must provide the nutrients and physical form required for the approved teaching organism. A sterilization cycle is effective only for the specified load, packaging, and validated equipment conditions. A displayed temperature alone does not prove every item in the load was sterilized. Process indicators, cycle records, and post-cycle inspection provide evidence, while an uninoculated control can reveal contamination. Different media may be damaged by excessive heating, so there is no universal cycle to invent here.

4. Procedure

  1. Confirm the selected medium, organism risk assessment, glassware-cleaning method, container fill limit, and validated sterilization cycle; obtain missing specifications from the approved local method.
  2. Under instructor supervision, measure and dissolve only the approved medium ingredients, label containers, and prepare a matching uninoculated control.
  3. Package glassware and medium for the specific validated load and record batch identification.
  4. Have trained staff operate the sterilizer and review time, pressure, temperature, and indicator records.
  5. After safe cooling, inspect the medium and containers, document quality-control outcomes, and store or discard as directed.

5. Calculation, graph, or images as appropriate

No nutrient mass or sterilizer setting is guessed. Assumed observations for practice only: an assumed cycle record is complete, an assumed indicator meets its criterion, and an assumed uninoculated control remains clear. These invented observations do not release a real load. Actual load validation and the release decision are determined by the authorized laboratory reviewer against the assigned validated sterilization method and recorded in the student record. This page decides nothing: it does not pass, fail, or release any load.

6. Result

Record actual observations here when the verified method and record are available. Safety note: Heat, pressure, glass, and microbial material create exposure risks. Only trained staff operate the sterilizer; use the institution’s containment and waste procedures.
