Study of Apparatus Used in Experimental Microbiology
1. Aim
To identify the purpose and key safety checks of apparatus used in a teaching microbiology laboratory. This is PCI Pharmaceutical Microbiology practical entry 1.
2. Requirements
Instructor-provided microscope, incubator, sterilizer or autoclave display, biosafety cabinet or clean bench illustration, pipette, loop, culture vessel, temperature indicator, and equipment log. Actual equipment model, controls, and operating ranges must be recorded here.
3. Theory
Microbiology equipment serves distinct jobs: a microscope visualizes cells, an incubator maintains specified growth conditions, a sterilizer treats defined loads, and a biosafety cabinet provides containment when correctly selected and operated. A clean bench protects a product but may not protect the operator, so the two must not be confused. Equipment labels, calibration status, and maintenance records matter more than memorized generic settings. Students should recognize the device and its limitations before any culture work.
4. Procedure
- Obtain the lab’s equipment inventory and the instructor-approved demonstration route.
- For each device, record its nameplate, purpose, basic controls, and current service or validation label.
- Observe an instructor demonstration of loading or setup without operating a sterilizer or cabinet independently.
- Match each apparatus to its role in observation, transfer, incubation, containment, or decontamination.
- Record one safety check and one common misuse for each, then return any movable items to storage.
5. Calculation, graph, or images as appropriate
| Observation | Actual teaching record |
|---|---|
| Equipment name and model | Record here |
| Primary function | Record here |
| Control or indicator | Record here |
| Service status | Record here |
| Safety check | Record here |
| Instructor-confirmed role | Record here |
No operating temperature or time is invented. Assumed observations for practice only: an assumed device has a temperature display and an assumed service label. Those invented features do not prove suitability for a real load. Actual settings and qualification must be recorded here.
6. Result
Record actual observations here when the verified method and record are available. Safety note: Do not open running sterilizers or handle live cultures as part of this survey. Use the laboratory risk assessment and instructor supervision.
