Biochemical Testing for Identification of Microorganisms

1. Aim

To interpret an instructor-supplied biochemical test panel for an approved teaching isolate and document the limits of identification. Isolate, panel, and decision database must be entered in the student record.

2. Requirements

Verified low-risk isolate record or de-identified panel dataset, manufacturer or course test instructions, positive and negative controls, incubation log, results sheet, identification key, protective equipment, and waste procedure. Exact reagents, incubation conditions, and interpretation thresholds are specified in your college lab manual and instructor's method sheet.

3. Theory

Biochemical tests reveal metabolic activities through colour changes, gas, substrate use, or other defined signals. A panel is interpreted as a pattern, not by one reaction. The result depends on a pure starting culture, validated media or kit, proper controls, and a database appropriate to the organism group. An unusual or inconsistent profile may require repeat testing or another identification method. Student practice should not be mistaken for clinical diagnosis or product release.

4. Procedure

  1. Confirm the approved isolate, purity evidence, panel instructions, and risk assessment; missing information is specified in your college lab manual and instructor's method sheet.
  2. Use instructor-provided results or a supervised low-risk panel prepared to the validated method.
  3. Record each reaction exactly as observed, including equivocal or invalid results.
  4. Check the positive, negative, and sterility controls before entering a profile into the approved key.
  5. Compare the profile with the reference database and report the level of confidence and any required follow-up.

5. Calculation, graph, or images as appropriate

No reaction concentration or incubation setting is fabricated. Assumed observations for practice only: an assumed panel has valid controls and an assumed profile matches one teaching reference. These invented data illustrate interpretation only; actual organism identity must be recorded here.

6. Result

The actual result and any identification or count must be recorded here until the verified method and observed record are available. Safety note: Use supplied datasets or instructor-approved low-risk isolates. Unknown and clinical samples require a separate institutional risk assessment and must not be cultured from this article.
