Study of Methods of Pure Culture Isolation

1. Aim

To interpret how an approved isolation method separates a mixed inoculum and to assess a candidate isolated colony. Organisms, media, and incubation are specified in your college lab manual and instructor's method sheet.

2. Requirements

Instructor-supplied plate images or approved low-risk teaching culture, labelled medium, sterile transfer tools, incubator record, isolated-colony images, protective equipment, and waste route. Organism identity, plate method, inoculum, and incubation conditions are specified in your college lab manual and instructor's method sheet.

3. Theory

A pure culture contains one intended microbial type, but an isolated-looking colony is only a candidate. Streaking progressively reduces the number of cells deposited so individual colonies can appear; spread and pour plates are other separation methods with different strengths. Colony morphology, a follow-up stain, and re-isolation can support purity. Unknown environmental cultures should not be handled as a beginner exercise. PCI asks for study of isolation methods, so a verified image sequence is suitable when live work is not locally approved.

4. Procedure

  1. Confirm the selected isolation method, organism risk assessment, and approved medium; missing details are specified in your college lab manual and instructor's method sheet.
  2. Examine a labelled sequence of plate images or observe an instructor demonstration.
  3. Identify the starting inoculation region, successive separation regions, and candidate isolated colonies.
  4. Record colony features and any areas showing mixed morphologies or contamination.
  5. Check the candidate against follow-up purity evidence provided by the instructor and write a conditional conclusion.

5. Calculation, graph, or images as appropriate

No culture dilution or incubation condition is invented. Assumed observations for practice only: an assumed final plate region contains well-separated colonies of one visible morphology. These invented observations suggest a candidate colony but do not prove pure culture. Actual purity must be recorded here.

6. Result

The actual result and any identification or count must be recorded here until the verified method and observed record are available. Safety note: Do not isolate unknown environmental or clinical organisms. Use approved teaching strains or images and follow instructor containment and disposal rules.
