Introduction to Biology Experiments
1. Aim
To orient students to the microscope, biological specimens, observation records, and safe handling used in the PCI Biology practical sequence. This is the first listed Biology practical entry.
2. Requirements
Compound light microscope, prepared plant and animal slides, blank slides, coverslips, observation notebook, labels, lens tissue, approved handling tools, and instructor-provided specimen list. Any stain or mountant for a demonstration is specified in your college lab manual and instructor's method sheet.
3. Theory
Biology practical work begins by separating what is observed from what is inferred. A microscope enlarges a small region but does not reveal every cell component; magnification and focus affect the visible detail. A biological drawing should show only structures actually seen, with a clear title, specimen identity, view, scale information when calibrated, and labels. Prepared slides are better for initial orientation because their tissue and stain are known. Unknown specimens require matched references and careful uncertainty notes.
4. Procedure
- Check the microscope, objective lenses, light source, slide stage, and clean optical surfaces without touching lenses with fingers.
- Record the specimen name or assigned code and note whether the slide is prepared, fresh, or a demonstration.
- Begin at the lowest objective, centre the field, focus gently, and progress to higher magnification only when needed.
- Record visible structures, sketch the field, and label only verified features.
- Compare the observation with the instructor reference, identify any artifact or uncertainty, and clean and return equipment.
5. Calculation, graph, or images as appropriate
| Observation | Actual teaching record |
|---|---|
| Microscope identification | Record here |
| Assigned slide or specimen | Record here |
| Objective and eyepiece labels | Record here |
| Visible structures | Record here |
| Sketch or labelled record | Record here |
| Instructor-confirmed identification | Record here |
Assumed values for practice only: assume an assumed eyepiece label of 10× and an assumed objective label of 4×. These invented instrument labels illustrate the calculation and are not readings from the student microscope.
- Illustrative total magnification = assumed 10 × assumed 4 = 40×.
- Actual magnification and any calibrated field size must be recorded here from the instrument used. No cell size is invented.
6. Result
The actual result and any identification or yield conclusion must be recorded here until the teaching method and observed record are confirmed. Safety note: Carry microscopes with both hands, handle glass carefully, and use only approved teaching specimens. Dispose of broken glass through the instructor.
