Preparation of Cascara Elixir BPC
1. Aim
To study the extraction and formulation stages of Cascara Elixir BPC and, where an authenticated formula and trained supervision are available, prepare a teaching-lab sample. Record the formula calculation, extract handling, final volume, and appearance. The applicable BPC edition and local method must be specified in your college lab manual and instructor's method sheet. [1, 2]
2. Requirements
The East Point College of Pharmacy manual lists cascara coarse powder, liquorice coarse powder, light magnesium oxide, saccharin sodium, flavoring oils, alcohol, glycerin, and purified water. The authentic BPC formula, ingredient identities and grades, quantities, alcohol strength, extraction method, target volume, and storage instructions must be specified in your college lab manual and instructor's method sheet. A supervised setup may require a balance, beakers, stirrer, covered extraction vessel, percolator, water bath, filter, calibrated final container, label, and batch sheet. Wear a laboratory coat, eye protection, and gloves. Keep alcohol and flavoring oils away from flames and ignition sources. [2, 3]
3. Theory
This elixir combines a plant extract with sweetening and flavoring ingredients. The teaching manual describes moistening and extracting cascara with liquorice and light magnesium oxide, concentrating the liquid extract, then adding separately prepared sweetener and flavoring portions. Glycerin contributes to the vehicle. A percolator passes liquid through a packed plant bed; the collected liquid is the percolate. Concentration removes some liquid, so the final volume must be adjusted only after the portions are combined. [2]
The manual states an aim of one batch size but its working formula appears scaled to a different final volume. Its intermediate quantities and alcohol strength must therefore be reconciled against an authenticated BPC source before students measure ingredients. Clarity and color alone do not demonstrate extract identity, strength, stability, or patient suitability. [2, 3]
4. Procedure
- Obtain the authenticated BPC formula or instructor-approved source, edition, assigned final volume, extraction instructions, and safety controls. Verify the batch scale independently. These details must be specified in your college lab manual and instructor's method sheet. If the formula cannot be reconciled, complete only the paper calculation and flowchart review. [2, 3]
- Inspect ingredient labels, protective equipment, ventilation, glassware, and the record sheet. Confirm the controls for hot water, alcohol, and plant-powder dust. The local approval and extraction setup must be specified in your college lab manual and instructor's method sheet. [2, 3]
- Calculate each ingredient quantity from the verified master formula and assigned final volume. Record the scale factor with its units and obtain an instructor check before weighing or measuring. [2]
- If wet-lab work is authorized, prepare the plant and magnesium oxide mixture according to the written method. Moisten it with the specified liquid and allow the prescribed extraction contact time. The actual liquid quantity and time must be specified in your college lab manual and instructor's method sheet. [2]
- Transfer the prepared material to the approved extraction apparatus and collect percolate according to the instructor's endpoint. Avoid running the bed dry or changing the solvent without approval. The extraction endpoint and percolate volume must be specified in your college lab manual and instructor's method sheet. [2]
- Concentrate the percolate using the supervised method. Control heat and prevent splashing. The permitted temperature and target concentrate volume must be specified in your college lab manual and instructor's method sheet. Do not heat a mixture containing alcohol over an open flame. [2]
- Prepare the sweetener portion and the separate flavoring portion with their specified solvents. Combine these with the cooled extract and glycerin in the verified order, mixing thoroughly. The ingredient amounts and order must be specified in your college lab manual and instructor's method sheet. [2]
- Transfer to a calibrated final container and add the approved vehicle to the assigned final volume. Record color, clarity, visible sediment, and actual volume. Filter only if the verified procedure calls for it. [2, 3]
- Label as a teaching sample with batch details and not for patient use. Follow the institution's storage and waste directions, which must be specified in your college lab manual and instructor's method sheet. [3]
5. Calculation, graph, or images as appropriate
| Record item | Verified master amount | Scaled amount | Actual observation |
|---|---|---|---|
| Cascara and other dry ingredients | Use authenticated formula | Record here | Record here |
| Extraction liquid and percolate | Use authenticated formula | Record here | Record here |
| Concentrated extract | Endpoint from authenticated method | Not applicable | Record here |
| Sweetener, flavors, alcohol, glycerin | Use authenticated formula | Record here | Record here |
| Final volume and appearance | Criteria record from the assigned current monograph or approved college method | Record here | Record here |
Worked scaling method: If an authenticated master formula specifies M grams of cascara for V mL of finished elixir, an assigned v mL batch needs M × v/V grams. For a batch one-tenth of the authenticated master volume, this is M/10. The actual M, V, v, and numerical result must be recorded here. Scale each other ingredient using its own mass or volume unit. The classroom manual's conflicting batch sizes must be resolved before any working formula is used. [2]
6. Result
State whether the work was a paper exercise or an authorized preparation. Record the reconciled formula, calculated quantities, collected extract, final volume, appearance, and deviations here. Do not claim BPC compliance or patient suitability without an authenticated source and required quality checks. [2, 3]
