Limit Test for Sulphates
1. Aim
To compare sulphate-derived turbidity in a prepared test solution with a matched reference standard. This is PCI Pharmaceutical Inorganic Chemistry limit-test exercise 1(b). The sample monograph, allowed sulphate limit, and selected variant are to be recorded from the assigned current monograph or approved college method in the student record. [1]
2. Requirements
Assigned sample, verified sulphate reference, approved barium-containing reagent and acidic medium, suitable water, matched comparison cylinders, pipettes, stirrer, timer, viewing background, coat, gloves, eye protection, and barium-containing waste vessel. Exact reagent identities, concentrations, volumes, timing, and sample preparation must be recorded here. [2]
3. Theory
Dissolved sulphate reacts with barium ions to form finely dispersed barium sulphate, seen as turbidity. A limit test compares the test mixture with a reference representing an allowed amount; it does not directly yield an exact concentration. The East Point manual presents more than one sulphate comparison sequence, so the instructor must choose one validated method and use it consistently for standard and test. Differences in acid, seeding, final volume, or development time change the appearance and invalidate a casual comparison. [2]
4. Procedure
- Identify the material monograph and write down its sample preparation and sulphate limit; missing information is to be recorded from the assigned current monograph or approved college method in the student record.
- Inspect and label matched blank, standard, and test cylinders.
- Prepare the prescribed sulphate standard and sample solutions with chloride- and sulphate-controlled water as specified.
- Add the approved acid and barium-containing reagent in the validated order to both preparations. Exact quantities are specified in your college lab manual and instructor's method sheet.
- Mix and allow the same development time under identical conditions.
- View the cylinders from the prescribed direction and compare test turbidity with standard; check the blank.
- Record pass, fail, or inconclusive only against the verified criterion and collect barium waste.
5. Calculation, graph, or images as appropriate
| Observation | Actual teaching record |
|---|---|
| Monograph and limit | Record here |
| Blank appearance | Record here |
| Reference turbidity | Record here |
| Test turbidity | Record here |
| Matched timing and decision | Record here |
Assumed observations for practice only: the blank is clear, the reference is moderately cloudy, and the test is less cloudy at the same time. If the verified monograph says the test must be no more turbid than the reference, this assumed case passes. No actual sample or universal limit is implied. This is a comparison exercise, so no invented numerical concentration is used.
6. Result
Report the actual comparison only after confirming the method, matched standard, blank, and specified acceptance criterion. If the method, standard, blank, or criterion is missing, make no pass/fail claim. The real pass/fail decision is made by the qualified reviewer against the assigned current monograph or approved method and recorded in the student record. This page decides nothing: it does not pass, fail, identify, or release any sample. Safety note: Barium reagents and acid can harm skin and eyes. Use teaching-lab supervision and collect barium-containing waste separately.
