Separation and Identification of Sulpha Drugs by TLC

sulpha drugs tlc
sulpha drugs tlc

1. Aim

To separate a supplied mixture of sulpha drugs on thin-layer plate and compare its spots with authenticated standards. The actual identities must be entered in the student record.

2. Requirements

Approved thin-layer plate, authenticated sulpha drugs standards, sample, validated mobile phase, chamber, spotting tools, visualization method, ruler, protective equipment, and waste plan. Follow the college method sheet for solvent composition, sample concentration, development distance, and visualization reagent. Acceptance criteria are to be recorded from the assigned current monograph or approved college method in the student record.

3. Theory

Planar chromatography separates compounds by their different interactions with a moving solvent and a stationary thin-layer plate. A retardation factor Rf is spot travel distance divided by solvent-front travel distance, both measured from the origin in the same run. Rf depends on system conditions and is not a universal identity constant. An unknown is supported by matching a reference run alongside it, including spot appearance and system suitability. Co-migration alone does not prove identity. A blank or control helps detect contamination.

4. Procedure

  1. Confirm the method, hazards, authenticated standards, and labels before handling solvent.
  2. Mark an origin and lane plan on the supplied diagram or supervised plate without damaging the stationary phase.
  3. Develop the standards and sample by the validated method, then mark the solvent front immediately.
  4. Use the approved visualization record and measure each spot centre and solvent-front distance from the origin.
  5. Calculate Rf for each valid spot and compare the unknown with standards from the same run.
  6. Report a supported match or unresolved identity, noting overlapping spots and method limitations.

Assumed values for calculation practice only: an assumed spot travels 3.0 cm and an assumed solvent front travels 6.0 cm from the same assumed origin. Assumed Rf = 3.0 / 6.0 = 0.50. Every distance and the Rf are invented. Actual solvent system, spots, and identities must be entered in the student record.

6. Output or final result

Report a labelled chromatogram and reference-supported interpretation for sulpha drugs. Actual identities: Record here.

Teaching-lab safety: Use instructor-approved solvent handling, ventilation, visualization, and chemical waste procedures. Do not infer a solvent recipe from assumed distances.

This page is a supervised-activity guide; it does not report a completed case.
